Clinuvel Pharmaceuticals is advancing SCENESSE, an already-approved drug for a rare blood disorder, into a pivotal Phase III trial for vitiligo treatment in patients with darker skin tones. The company released topline results from its Phase IIb study, CUV105, in December 2026, with efficacy and safety data to follow in the coming months. This represents a material regulatory milestone for a company seeking to expand its product portfolio beyond its core orphan indication.

The CUV105 study enrolled 210 patients globally to evaluate SCENESSE combined with narrowband UVB phototherapy against the current standard of care, which is narrowband UVB monotherapy. The trial measured efficacy using validated vitiligo scoring tools, with the primary endpoint being the proportion of patients achieving at least 50 percent improvement in total body surface area repigmentation and a secondary endpoint of at least 75 percent improvement on the face. These specific metrics are clinically relevant because facial and visible repigmentation carries particular psychological and quality-of-life significance for patients, especially in populations where vitiligo stigma remains acute.

The regulatory path forward has cleared a significant hurdle. The European Medicines Agency reviewed the protocol for the pivotal Phase III study, CUV107, in April 2026 and provided feedback on trial design. Clinuvel has incorporated this guidance, with CUV107 planned to begin enrollment in November 2026 and run for a 12-month recruitment window across 300 patients with darker skin types. Regulatory alignment at the protocol stage reduces the risk of late-stage trial redesign, which can delay approvals by years.

For investors, the expansion into vitiligo addresses a large unmet medical need. Vitiligo affects an estimated 1 to 3 percent of the global population, with darker skin types experiencing greater visual impact and psychological burden. Current treatment options remain limited and inconsistent, making SCENESSE a potentially meaningful therapeutic addition if efficacy holds across larger patient populations. The company already has manufacturing and regulatory experience with SCENESSE for erythropoietic protoporphyria, which should facilitate faster development and potential commercialization in vitiligo.

Case reports published ahead of the full CUV105 readout showed visible repigmentation in treated patients at 20 weeks, and earlier Phase II data suggested that repigmentation gains persisted during observation periods off treatment. These signals support the biological plausibility of the mechanism, though full efficacy and safety data from CUV105 will be determinative of clinical benefit and label scope.
